研究概要

Co14a6遺伝子ノックアウトマウスのヘテロ接合体およびホモ接合体を得ることができた。これらのマウスにおいてCo14a6遺伝子のRNA転写、タンパク翻訳活性について詳細に検索した。その結果、転写は認められず、遺伝子発現停止実験が成功したことを意味した。ノックアウトマウスと正常マウスの間の形質変化があるかどうかの検索を進めたところ、現在に顕著な差異は認めていない。
α鎖特異抗体を用いて、α1/α2、α3/α4/α5とα5/α6の三種の分子構成があることを特異抗体を用いた免疫二重染色法によって明らかにした。特に腎糸球体と肺胞基底膜はα3/α4/α5、皮膚および腎ボーマン嚢基底膜はα5/α6分子で構成され、これらの分子を含む超分子会合体でつくられる基底膜とその臓器における機能との関係がありそうである。さらに責任遺伝子がCOL4A5であるアルポート症候群の解析によって、ひとつのα鎖が変異を示すと他のα鎖が細胞内で分解されることから、上記のα3/α4/α5とα5/α6の構成分子が存在することが再確認された。
アルポート症候群に食道下部平滑筋症が合併する大変稀な症例を日本国内で見つけだし、詳細な解析を実施した。その結果、欠失部分はCOL4A5とCOL4A6上流域17kbに及ぶことがわかった。欠失周辺にはトポイソメラーゼコンセンサス配列が存在し、これらは大きな欠失の修復に関与したことが推定された。
