腎臓膜輸送体異常症の分子細胞生物学的研究

研究概要	1.輸送体の調節機構の解明蛋白レベルでの検討としては、AQP3は水チャネルのみならず、中性小分子も通過させるが、水と小分子が同一のporeを通るか否かは不明であった。水銀剤による阻害実験により両者は同一のporeを通ることが示された。さらにAQP2とAQP3のpore構造も同じであることが推測され、AQPファミリーの構造と機能の関連に示唆を得ることができた。細胞レベルでの検討としては、AQP2がvasopressinの刺激によって細胞内貯蔵部位の小胞より細胞膜表面へsortingされる現象を調べた。AQP2のC端近くに存在するA-kinaseリン酸化部位のリン酸化が引き金となって、sortingが起きることを明らかにした。輸送体蛋白の合成の調節については、核での遺伝子の転写制御についての検討を行なった。高浸透圧とcAMPによってAQP2,CIC-K1の転写が亢進することを示した。 2.輸送体の調節異常と病態の関連の明確化腎性尿崩症、Dent病、Liddle症候群において、全国の施設の協力を得て、患者DNAの解析を行い、多くの遺伝子変異を同定した。また疾患モデルを作ることも大切であり、CIC-K1,CIC-K2,CIC-3のノックアウトマウスを作成中である。またいくつかの輸送体については、その機能が過剰になった病態を調べるため、トランスジェニックマウスを作成中である。 3.腎臓尿細管特異的発現機構の解明尿細管セグメント特異的発現機構を明らかにするために、AQP2,CIC-K1,CIC-K2の遺伝子上流を用いて、発現、非発現の培養細胞においてin vitro reporter gene assayを行ない、臓器特異的cis elementを同定した。またこの遺伝子上流を組こんだreporter geneを導入したトランスジェニックマウスを作成中である。
