研究概要	AQPの構造に関しては、AQP3は、阻害剤・変異導入の解析によりAQP1と同じ構造であることが分かり、さらに酵母の発現系を用いたAQP2の検討で、第4、5膜貫通部を中心とした部分が水通過のporeを形成していると考えられた。AQP2については、DKAによる燐酸化はチャネルの透過選択性を変化させないことを示した。腎性尿崩症、Dent病、Liddle症候群の患者について、全国より依頼を受けて、患者のDNAのシークエンスを行ない、いくつかの新しい変異を同定した。日本の腎性尿崩症の11家系を解析し、AQP2変異を4家系で認めている。確認のできた変異について発現実験で機能を確認したが、機能不全に陥っている変異が発見された。さらにAQP2変異は通常、劣性の遺伝形式をとるが、優勢の形式を取る例を見つけており、その機序は現在検討中である。 ClC-K1についてはその性質・存在部位から尿濃縮に関与していると推察していたが、確証を得るためにノックアウトマウスを作成した。ClC-K1ノックアウトマウスは正常に生まれてくるが、多尿を示し、バゾプレッシン投与にても尿浸透圧は上昇せず、腎性尿崩症であった。従ってClC-K1の生理的役割は確定した。現在腎性尿崩症患者でClIC-K1遺伝子異常の検索中である。ClC-5の遺伝子異常でDent病になるが、我々も5家系のDent病患者で解析を行い4家系でClC-5の遺伝子異常を見つけている。免疫染色により、ClC-5蛋白がH-pumpと細胞内小胞(endosome)に共存している像を認めた。従ってClC-5はendocytosisに必須のClチャネルであり、その障害で各種臨床症状が出現すると予想される。腎臓尿細管特異的発現機構については腎特異的なNaK2Cl輸送体の上流部分2kbを組み込んだトランスジェニックマウスを作成し、現在その組織分布を検討中である。
