体液・循環調節に関与するペプチドホルモン受容体に関する比較分子生物学的研究

研究概要	特殊な浸透圧及びpH環境に適応している魚類を中心とする比較分子生物学的なアプローチにより,循環系の恒常性維持機構の解明に新視点を導入するべく研究を続け,本年度は以下の成果を得た。1)pH3.5という驚異的な湖水に適応している恐山ウグイの酸性適応機構の解明から明らかになった重炭酸イオン輸送体の新規哺乳類ホモログの一つが,脈絡叢にのみ高発現し,脳脊髄液のpH及びイオン調節に重要な働きをしていることを明らかにした。2)pHセンサーの可能性がある分子で私たちが見つけたIg-Heptaのプロセッシング部位を同定した。3)Na, K-ATPaseのパートナーで細胞にとって必須の新規Kチャネルのドミナントネガティブ変異体を作成し,機能解析系を立ち上げた。4)Na利尿ペプチド(NP)ファミリーが魚類の広塩性を支配する重要な内分泌系であることを明らかにしたのに引き続き,ヌタウナギ,チョウザメ,ハイギョなど系統進化上興味深い動物でNPを同定した。5)A型及びC型NP(ANP, CNP)がそれぞれ海水及び淡水ウナギでコルチゾルの分泌を促進することを明らかにするとともに,コルチゾル生合成系に関わる酵素をウナギでクローニングした。6)ウナギで浸透圧調節に関わる可能性が高いグアニリンを3種同定した。7)広塩性魚類ティラピアで2種のクロライドチャンネルと細胞間イオン輸送に関わる5種のクローディンを同定し,浸透圧適応機構の理解に欠くことのできないタイトジャンクションにおける選択的イオン輸送解明の足場を築いた。
