グロビン遺伝子群の多型を用いた日本人集団の分子人類遺伝学的研究

研究概要	本年度はアフリカ、地中海沿岸域から中近東、インド、東南アジア、中国にかけ高い頻度で見られるサラセミアにおけるβグロビン遺伝子の変異と多型とに注目し、目本人の起源を検討した。なかでも北方起源の可能性を見るために中国および韓国の、また南方起源の可能性を見るためにタイ、マレーシア、インドネシアのサラセミアの変異と、変異に連鎖したグロビン遺伝子群の複数の多型の組み合わせからなるパターン(ハプロタイプ)と、遺伝子内の多型の組み合わせ(フレームワーク)を検討した。その結果、各集団間に共通に見られる変異としては、日本人とタイ人、マレーシア人、インドネシア人ではIVS-2 nt654(C-T)、codons 41/42 4bp欠失の2種類であり、日本人と韓国人とはcodons 41/42 4bp欠失、codon 121(G-T)、IVS-2 nt1(G-A)である。しかし山口大学の大庭や川崎医科大学の原野により、韓国人で見られるIVS-1 nt130(G-C)、initiation codon(T-G)またcodon 17(A-T)の変異が日本人でも見出されており、これらも加えると6種類の変異を共有することとなる。多型のパターンつまりハプロタイプ及びフレームワークを検討した結果、東南アジアでも見られる日本人の変異は中国人由来と考えられた。一方韓国人および中国人にも見られる変異は同様に中国人由来と考えられた。韓国人および日本人でのみ見られる変異の起源は同定できなかったが、今後多型を詳細に検討することにより推測が可能と考えられた。以上をまとめると、日本人の起源をサラセミア変異および多型から検討した結果、中国・韓国人起源が主であり、東南アジア人起源の可能性は低いものと考えられた。
