硝酸呼吸系に含まれる銅タンパク質の構造と機能の研究

研究概要

本研究では、Alcaligenes xylosoxidans GIFU 1051からの青色NIR(Axg-NIR)遺伝子のクローン化と結晶構造解析を行ない、既に報告のある亜硝酸還元酵素(NIR)と比較した。
Axg-NIRのDNA塩基配列から推定されるAxg-NIRのアミノ酸配列を他のNIRの配列と比較した結果、同じ青色NIRであるAlcaligenes xylosoxidans NCIB 11015(Axn-NIR)およびPseudomonas aureofaciensのNIRは共に77%、緑色NIRであるAlcaligenes faecalis S-6,・Achromobacter cycloclastes IAM1013のNIRとそれぞれ67%、70%の高い相同性を有していることが明らかになり、同様の高次構造を取ることが示唆された。また、2つの銅の配位残基であるType1銅の2His,Met,CysおよびType2銅の3Hisはすべて完全に保存されており、銅中心の構造も類似しているものと考えられる。
酸化型Axg-NIRのX線結晶構造解析(2A^^.分解能)では、既に報告のあるAxn-NIR(3A^^.分解能)よりも精度のよいデータが得られた。それによると、Type1銅は2His,Met,Cysの4残基が配列した歪んだ四面体構造であり、同じ配位残基を持つが、緑色NIRの銅周りのように歪んだ平面四配位とは異なっていた。Type2銅部位は、今までに報告のあるNIRと同様で、3His残基と水分子が配位した歪んだ四面体構造であった。また還元型では、酸化型と比べてType1銅部位は変化なかったが、Type2銅部位の水分子が外れて三配位構造であった。
