転写基本因子群及び相互作用因子群の持つ新しい構造モティーフの三次構造解析

研究概要	転写基本因子群が多様な転写調節因子群に相互作用し、転写調節反応に関わっていくためには、これらが特徴的な構造を有していることが必要である。そこで、転写基本因子群の機能の基盤になる分子構造を三次元レベルで解析するために、それらが有する特徴的な構造を一次構造から見い出し、転写調節における分子間相互作用の特異性を理解し、転写調節における転写基本因子群の機能的役割を明らかにすることを目的として解析を進めた。 1)新しいタイプのクロマチン転写因子群の単離 TATAボックス結合因子TFIIDや転写基本因子群と相互作用する新しいタイプのクロマチン転写因子群を単離することを目的に蛋白質-蛋白質間相互作用活性を利用するtwo‐hybrid法や蛋白質クロマトグラフィー法を用いた。TFIIDサブユニットや転写基本因子群の様々な構造ドメインに相互作用する因子を単離し、その結合特異性、細胞内在局性、発現分布、転写活性化能などに解析した。その中には細胞周期やシグナル情報伝達システムに関わる因子、更にヒストンアセチルトランスフェラーゼ活性あるいはクロマチン構造変換活性を有する新しい因子などを含む数多くの未知因子が含まれていた。 2)単離したクロマチン転写因子群の構造解析用大量調整法の確立と結晶化これらの因子群の構造-活性相関の解析を三次元レベルで行うために大腸菌発現系で大量発現させ、精製することを試みた。様々な因子について大量調整法の確立を行い、結晶化条件の検討を加えている。
