巨核球成熟と血小板放出機序の細胞分子生物学的解析

研究概要	1)TPOによって巨核球が成熟分化することが明らかになったが、巨核球の多倍体化や血小板放出の分子機序に関しては不明のことが多い。そこでTPOによる多核球の多倍体化のメカニズムを解明する目的で、骨髄細胞から巨核球を単離しTPOを投与後、多倍体化する巨核球を細胞生物学的に解析した。その結果、株化された細胞で言われているような細胞分裂(M期)をスキップしてDNA合成のみが反復されるメカニズムではなく、巨核球はM期に入りprophase、metaphase、anaphase Aまでは進行するものの、姉妹染色体が極まで充分引かれないために核膜が複製した染色体を全て包括してしまう為に起きることが判明した。 2)TPO、EPO、IL3等の造血因子によって従来アポトーシスのシグナル伝達に重要と考えられていたJNKやp38が一過性に活性化されることが明らかになったが、その造血細胞に於ける機能は不明のままであった。そこで今回はその作用を解析した。その結果、細胞成熟が誘導される細胞株では、ERKは余り関与していないがJNKやp38が細胞成熟に重要な役割を担っていることが判明した。しかし依存して増殖する細胞株に於いては細胞増殖に重要な機能を果たしていることも判明した。従って細胞の状態によって役割分担が異なる可能性が判明した。 3)血小板放出(PPF)にはTPOは全く関与しておらず、PPF形成のメカニズムに関しては不明なことが多い。そこでPPFアッセイ系を確立し外来因子の存在の可能性を調べた。その結果、PPFが促進される因子が血小板減少症患者血しょうに存在すること、およびある細胞上清にも同じ分子量分画にPPF促進活性が見い出されin vivoでも即応する因子を同定した。
