研究概要

本研究は大別して、IL-3受容体の遺伝子導入(「卜」)マウスの交配・系統維持、同「卜」マウスの造血幹細胞/前駆細胞の機能アッセイ、同「卜」マウスの諸臓器におけるpleiotropismの解析、等から構成されており、以下に実績の概略を示す。
1.交配と系統維持(平林・梅村・井上):既に作製されたヒトIL-3受容体の各サブユニット遺伝子導入マウスのうち、α鎖とβ鎖のマウスの交配により双方の遺伝子導入マウスを得、以下の解析に供した。 尚、それぞれの「ト」マウスはホモ化して維持する計画だったが、現在までにホモ個体を確認するには至らなかった。また、IL-3分泌マウスとの交配実験については、まとまった結果を得るに至らなかった。
2.造血幹細胞/前駆細胞の機能アッセイ(平林・横田・井上):(1)α鎖マウスの遺伝子発現:新規市販品の2次抗体を用いることで、骨髄・脾・胸腺細胞の導入遺伝子発現分画をセルソーターで従来より明瞭に示すことが出来た。(2)αβマウス骨髄細胞の反応性:α鎖とβ鎖のマウスの交配により作成した双方の遺伝子導入マウス骨髄細胞は、ヒトIL-3に農度依存性に反応してln vitroコロニーを形成した。形成されたコロニーの約1/4はcompactな形態を示し、表面分化抗原をセルソーターで解析したところ、B220及びCD19が陽性のB細胞系列であることが解った。(3)compact colonyの性状:このcompact colonyはヒトIL-3添加によって二次性コロニーを形成するが、マウスIL-3添加では全く増殖しない。ヒトIL-3にマウスIL-7を添加して二次性コロニーを形成させるとCD43陽性分画が減少し分化傾向が認められた。
3.諸臓器におけるpleiotropismの解析(高木・平林・井上):受容体「卜」マウスの諸臓器での導入遺伝子発現をRT-PCR等で観察し、小脳や精巣等にも導入遺伝子の増幅産物を認めた。
尚,必要な備品類はほぼ揃っており、「卜」マウスの選別のための遺伝型決定などの効率向上のため卓上微量高速遠心機を充当した。また頭目的のためにアルバイトも依頼した。
