研究概要

1.日本、ロシア、中国、香港、マレーシア、ブラジル、コロンビアなどの胃癌症例について5-11%の頻度でEBV関連胃癌を確認した。
2.EBV関連胃癌の内視鏡的特徴はリンパ球浸潤を反映して、表面に陥没を有し境界不明瞭に隆起し、その背景胃粘膜は、萎縮性変化に富んでいた。
3.粘膜内のEBV関連胃癌について癌の発生の極めて早期にEBV感染が成立していることを明らかにした。CD44のvariant3-5,6が特異的に強く発現していることを見出した。
4.胃癌組織由来のEBV感染上皮系細胞株3株が樹立された。細胞株は軟寒天内でコロニー形成能があり、SCIDマウスで腫瘍を形成した。
5.EBVのエンベロープタンパクgp85のプロモーター領域の欠失組換えEBV(△TK/gp85-EBV)を作成した。本ウイルスはBリンパ球への感染性を失っていたが、上皮細胞への感染性を保持していた。
6.膿胸関連リンパ腫(PAL)は全例がEBV陽性であり、高率にp53遺伝子変異が見られた。NK細胞性鼻腔リンパ腫とEBV陽性との関連が強く示された。
7.LMP1の機能と発現制御に関して(1)LMP1発現によるTGF-β1への細胞の感受性低下が腫瘍形成に重要である。(2)CD40単独刺激によるLMP1発現の特異的制御を明らかにした。
8.LMP1はJurkat T細胞においてはBcl-2の発現増強を介さずアポトーシスに抵抗性を獲得させた。BHRF1発現胎児由来小腸上皮細胞ではTNFαや抗Fas抗体依存性アポトーシスに抵抗性を示した。
