研究概要	環境因子の発がんリスク評価のための信頼に答えうる基礎的データを作ることを目的とし、次の如くの結果を得た。1)2-amino-1-methyl-6-phenylimidazo[5,6-b]pyridine(PhIP)投与ラット大腸におけるaberrant crypt foci(ACF)の発生は平坦-立ち上がり曲線を示し、PhIPの発がん性には無作用量の存在が強く示唆された。2)2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline(MeIQx)によるラット肝のglutathione S-transferase placental form(GST-P)陽性細胞巣の発生には系統差や経胎盤、経乳による影響はみられなかったが、20%食餌制限はその発生を減少させた。3)非遺伝毒性発がん物質であるdieldrinによるGST-P陽性細胞巣の発生はU字型曲線傾向を示し、その肝発がん性には閾値が存在する可能性が示唆された。4)dimethylhydrazineのラット肝に対するイニシエーション活性の低用量加算効果を認めた。5)各種トランスジェニックあるいはノックアウト動物の変異原活性、発がん感受性を検討した。低用量発がんリスク評価に対する有用性にはさらなる追究を要する。6)胎児および新生児において8-hydroxyguanine(8-OHG)レベルを検索すると、8-OHGは出生後、種々の組織で検出され、日齢が増えるにつれて増加した。7)新生児はミルクを介してMeIQxに曝露され、肝にMeIQx-DNA付加体が検出された。8)がん関連遺伝子変異の高感度検出法としてPeptide nucleic acid法、PLACE-SSCP法を検討した。
