ヒト肺癌の発生・進展過程における遺伝子変化

研究概要	肺癌の分子病態のより多面的かつ包括的な解明に努めることを目指した研究を進め、以下の結果を得た。 (1)肺癌に高頻度にみられる17p13.3欠失のセントロメア側共通欠失領域内に検出したホモ欠失のphysical map作成及び候補癌抑制遺伝子の単離・同定を試み、G2チェックポイント関連遺伝子が含まれていることを明らかにした。また、染色体の分配制御にかかわるmitotic checkpointの異常を約40%の肺癌細胞株に検出した。さらに、MAD1遺伝子に体細胞変異が低頻度ながら検出されることを、初めて明らかにした。Vogelsteinらによる大腸癌におけるBub1遺伝子変異の報告を除き、唯一のmitotic checkpoint関連遺伝子変異の報告である。 (2)17p13.3のテロメア側共通欠失領域に存在する新規ヒトserpin遺伝子(CASPIN/PEDF)ついて、我々が樹立した高転移性ヒト肺癌細胞亜株H460Luへの遺伝子導入による検討を加えた結果、有意な血行性転移抑制能を持つことが明らかとなった。さらに、肺癌患者組織における変異の検索及び発現量の変化について検討を進めつつある。 (3)酸化DNA障害の軽減に関与するglutathione peroxydase(GPX)遺伝子や、喫煙の習慣性に関与する可能性が考えられるdopamine D4 receptor (DRD4)遺伝子を対象とし、其々の遺伝子多形の肺癌発症或いは喫煙習慣に対する危険寄与度について検討を加えたが、有意な相関は検出されなかった。
