新規血管新生抑制因子の分離とこれを用いたがん転移阻止の試み

研究概要

血管新生なしにはがんは増殖しえず転移巣も形成されない。したがって、血管新生制御のしくみを解き明かすことは制がん上の重要命題である。本研究の目的は、2種の新しいアプローチにより血管新生を制御する新しい因子の分離を試みることである。一つは″Antisense Display法″による内皮細胞増殖関連遺伝子のスクリーニングで、もう一つは心臓が産生する血管新生抑制因子の分離である。
1.Antisense Display法による新規血管新生関連遺伝子のスクリーニング
(1)10merアンチセンスレパートリーをサブグループに分け、ヒト微小血管内皮細胞の増殖を指標にスクリーニングを行い、最終的に内皮細胞増殖を抑制する単一のアンチセンス分子種を同定した。
(2)同定されたアンチセンス配列に対応するセンスプライマーとアンカー型オリゴ(dT)プライマーを用い内皮細胞ポリA^+RNAを鋳型にRT-PCR法を行い候補cDNAを3種分離した。DNA配列データベースでの相同性検索の結果、前者はミトコンドリアNADH-ubiquinone oxidoreductase chain3をコードするcDNAであることが判明した。また、後2者は新規遺伝子で、clone30は酵母のMIC1 cDNAと相同性を示し、clone32はマウスのNck interacting kinase(NIK) cDNAと高い相同性を示した。
(3)Clone 2、30、32の配列に特異的な16-18塩基のアンチセンスオリゴヌクレオチドおよび対照のセンスオリゴヌクレオチドをヒト微小血管内皮細胞の培養液に添加し、内皮細胞の増殖阻害を指標に機能検定を行った。その結果、clone2と30に相補的な長鎖アンチセンスがDNA合成を阻害した。Clone32に対するアンチセンスは、センスオリゴヌクレオチドと同様内皮細胞DNA合成に影響を及ぼさなかった。
2.新規血管新生抑制因子の分離・精製
(1)ヒト微小血管内皮細胞の培養液にウシ心臓抽出物を添加すると、内皮細胞の増殖が阻害されることが見い出された。心臓抽出物を種々のカラムクロマトグラフィーで分画し、各画分の内皮細胞増殖阻害活性を測定した結果、いくつかの異なる画分が阻害活性を示した。
(2)当該画分を、ヒト微小血管内皮細胞と平滑筋細胞の培養に添加したところ、少なくとも2種の画分が内皮細胞の増殖を特異的を阻害した。
3.次年度以降の研究計画で血管新生制御による癌転移阻止の可能性を追求するためのモデルとして予定している胃癌および大腸癌同所移植転移マウスモデルで、原発巣と転移巣形成のtime courseを決定した。
