細胞内増殖機構における蛋白質リン酸化酵素PKNの機能に関する研究

研究概要

本研究においては、PKNと直接的、間接的に相互作用することによりシグナリングを制御している分子の同定を種々の方法を用いて行い、既知のシグナリング分子との関連も含めて、複雑なシグナリングネットワークの解明の一助とすることを目指した。以下に、得られた結果の概略を記す。
1.PKNと相互作用する蛋白質の解析
1)ビメンチン
ニューロフィラメントの他に、普遍的に存在する中間径フィラメントであるビメンチンともPKNが結合かつリン酸化し、重合・脱重合を制御していることから、PKNがこれら中間径フィラメントのダイナミックな動きを制御している可能性が示唆された。
2)PCD17
卵巣癌、子宮癌や乳癌を伴う亜急性小脳変性症(Paraneoplastic cerebellar degeneration)の自己抗体により認識される抗原としてクローニングされているPCD17が、PKNと特異的に結合することを見出した。PCD17はその一次構造上の特徴から転写因子としての機能が予測されていたが、いわゆるOne hybrid systemにより、これが転写活性制御因子であることを実験的に示唆した。このことより、PKNが核内においても何らかの機能を有することが期待された。
2.PKN基質蛋白質の解析
PKNの生理的機能を知る上で、その特異的基質を検索することはきわめて重要である。我々は、種々の組織抽出液よりPKNが特異的にリン酸化する蛋白質の同定を試みている。今回は、ラット肝臓よりPKN基質となりうる新規の蛋白質を精製し、そのcDNAをクローニングし解析した。本蛋白質(MIPP65)はミトコンドリアNADH ubiquinone oxidoreductase 9 KD a subunit precursorと有意な相同性を示し、動物細胞中で発現させるとミトコンドリアに局在する事が明らかとなった。
