研究概要

微生物由来ならびに海洋生物由来生産物質から各標的に対する活性物質を精製単離、構造決定し、1および2に記載する物質を得た。前臨床研究の一部として3を行なった。
1.新規制癌物質の探索を各標的について実施し以下の物質を得た。培養細胞に対する直接細胞毒性を指標として海洋生物生産物より13種のRitterazine誘導体、蛋白質脱リン酸化酵素阻害作用を示す5種のCalyculin誘導体、DNAポリメラーゼβ阻害作用を示すYaurosponginA、チロシンキナーゼ阻害作用を示すTauroacidin A,B、アポトーシス誘導物質としてJolkinolides、微生物生産物よりPhosmidosine、微小管脱重合阻害物質として植物成分よりTaxuspine Dを得た。
2.補助療法に有用な物質として以下の物質を得た。多剤耐性克服物質として植物成分よりTaxuspine Cを、微生物由来物質よりAndrastin Aを得た。TGF-β様活性物質として微生物由来の新規物質Diphetepeptin,TGF-β活性調節物質としてBaffilomycin類縁体を得た。サイトカイン活性修飾物質として微生物由来IL-6活性阻害物質Madindoline A,B、海洋生物よりNorzoanthamineを得た。また、各種のサイトカインの活性修飾物質として微生物由来物質Cytoxazoneを得た。細胞接着分子VCAM-1阻害物質としてHalichlorineを得た。
今後、これらの物質についてその作用機構、制癌活性等を検討し有用物質を選択する予定である。
3.制癌物質の重要な限界因子である消化管毒性に軽減作用を示す低分子サイトカイン産生調節物質Conageninの作用を消化管細胞における各種サイトカインのmRNAを検出することによって追求し、本物質が制癌物質によって誘導される消化管毒性の原因となる炎症性サイトカインTNFαのmRNAの発現を抑制し、抗炎症性サイトカインIL-10のmRNAの発現を亢進することを明らかにした。
