肝胆道腫瘍遺伝子治療のための新世代アデノウイルスベクターの開発

研究概要

我々は以前より自殺遺伝子を発見する組換えアデノウイルスを用いた遺伝子治療の基礎研究を行ってきた。自殺遺伝子療法は癌細胞特異的に遺伝子導入効率を高めることができれば、さまざまな癌種において最も確かな効果が期待できる。しかし癌細胞が活性化された薬剤そのものに耐性である場合が存在すること(Cytosine Deaminase/5FCの系においては活性化された5FUに耐性をもつ癌細胞がある)が問題となっていた。5FUは固形癌の治療に汎用されるが、単独での奏功率は低く耐性が多い。哺乳類細胞では5-FUは複数の経路で代謝され、主にFUTPとFdUMPが細胞毒性を発揮するが、5FUがFUMPになる最初の反応が1つの律速段階となっている可能性がある。我々は大腸菌にUracilをUMPに変換するUracil phosphoribosyltransferase(UPRT)が存在することに着目し、大腸菌UPRT遺伝子を組み換えアデノウイルスベクターで各種消化器癌細胞に発現させ5FUを効率よくFUMPに変換することで、5FUに対する耐性を克服できないか検討したところ、UPRT遺伝子を導入された細胞では5FUに対する感受性が6.3-100倍上昇した。ヒト癌を皮下に移植した動物モデルを用いた検討では、5FUの全身投与のみでは腫瘍は縮小しなかったが、ウイルスと5-FU投与による治療群では著明な腫瘍縮小効果が得られた。大腸菌UPRT遺伝子をを導入効率の高いアデノウイルスベクターで癌に導入するまったく新しい方法で、5-FUの耐性を克服できる可能性がある。
