研究概要

IL-15はIL-2Rβ、γ鎖をレセプターに使用するサイトカインでIL-2と同様の生物活性を示し、NK細胞、αβT細胞及びB細胞の増殖活性化因子として働く。活性化T細胞によって産生されるIL-2とは異なり、マクロファージや上皮細胞が産生されることが特徴である。今回、IL-15の抗腫瘍活性をIL-15遺伝子導入Meth A線維肉腫とBALB/Cマウスの系で検討した。
我々はIL-15mRNAにはalternative splicingによって少なくとも4種類のアイソフォームが存在することを明らかにし、その中で、長いエクソン5を持つアイソフォームが高い蛋白への翻訳効率を有することを見いだした。翻訳効率の高いcDNAと翻訳効率の低いcDNA導入Meth A細胞からRNAを抽出してIL-15mRNAの発現を確認し、さらに培養上清中にIL-2/IL-15依存性CTLL-2細胞の増殖活性を確認した。この2種類のIL-15cDNA導入Meth AトランスフェタントをBALB/Cマウスの皮下に投与して、腫瘍の大きさを経時的に測定したところ、IL-15高産生Meth Aトランスフェクタントで増殖抑制が最も顕著であった。ヌードマウスでは2つのトランスフェクタントとともに増殖は同様に認められ、またCD8抗体投与マウスではIL-15高産生Meth Aトランスフェクタントの増殖抑制を認められなかった。以上の結果より高翻訳のIL-15cDNAアイソフォームを用いた遺伝子導入は有効な抗腫瘍効果をもたらすことが明らかとなった。
