腫瘍内休止期細胞への効果を加味した中性子捕促化合物の効果投与様式の探究

研究概要

中性子線照射に対する休止期細胞(Q細胞)の感受性及び、中性子捕捉化合物の腫瘍内分布を改善させるための併用処置に関し、以下の諸点を明らかにした。
1.中性子捕捉化合物不投与下での中性子照射に対する腫瘍内のQ細胞の感受性は全腫瘍細胞(P+Q細胞)よりも低いがγ線照射時程の差はなく、従って、γ線照射時と比べた相対的生物学的効果比(RBE)は、Q細胞の方がP+Q細胞よりも大きい。また、低Cd比中性子線の照射の方がこのRBEが大きくなる傾向があった。
2.中性子捕捉化合物投与下では、Q細胞もP+Q細胞も感受性が高まるが、高Cd比中性子線照射で著しい。P+Q細胞においてはBPAが、Q細胞においてはBSHが顕著に感受性を高めたが、MRI造影剤のGd化合物にはそのままではほとんど感受性増強作用がなかった。
3.中性子捕捉化合物腹腔内投与時の併用処置としてはニコチンアミドの前投与が、Q細胞の感受性の増強には(低温度)温熱処置の併用や生体還元物質のtirapazamineの併用投与が有効と考えられた。
本年明らかになった事項を基にして、さらに中性子照射後の腫瘍内P+Q細胞並びにQ細胞における(再)酸素化及び再分布現象を明らかにする事により、不十分量の中性子捕捉化合物しか腫瘍に分布させ得なかった症例や、腫瘍の存在部位と照射設備との都合により不十分量の中性子しか照射させ得なかった症例に対し、再発予防を目的とするγ線あるいはX線照射を追加する際のより効果的な照射タイミングを明らかにする事を今後の目的としたい。
