ヒトがん細胞のアポトーシスを誘発するオピオイド関連受容体の分子生物学的研究

研究概要	オピオイド化合物であるブプレノルフィンが、ヒトがん細胞由来のA549細胞に対して細胞致死作用を示したこと、ならびに核の断片化および凝縮を主徴とするアポトーシスの形態をとる細胞変化を誘発させたという昨年度の報告に基づいて、本研究ではさらに詳細な検討を加えた。まず、細胞周期および核の形態変化をフローサイトメトリーを用いて解析したところ、ブプレノルフィンを処置したA549細胞では、薬物未処置の対照群に比し、G2/M期における顕著な細胞数の減少とG1期における2nよりも小さいの核の断片の存在が確認された。さらに、同様に処理した細胞から抽出したDNAを電気永動したところDNAの断片化すなわちラダーリング現象が観察された。以上の成績から、ブプレノルフィンによるがん細胞致死作用はアポトーシス誘発に起因することが強く示唆された。アポトーシスの発現にカスパーゼ(ICE/CED-3)ファミリープロテアーゼが関与することが知られているが、カスパーゼ1(ICE)阻害剤であるZ-Asp-CH2-DCBの併用処置によってブプレノルフィンによる効果は完全に抑制された。これらは、ブプレノルフィンのアポトーシス誘発作用においてもカスパーゼ1を介する機構が重要な役割を果たしていることを意味している。一方、鎮痛薬の抗がん活性の構造活性相関を検討する過程で、末梢性作用型の合成オピオイド化合物であるロペラミドがブプレノルフィンよりも強い活性を有することを見いだした。その薬物のがん細胞に対する作用特性の詳細については現在検討中である。
