抗腫瘍免疫におけるT細胞活性化の制御

研究概要

T細胞の活性化は、TCRを介する抗原認識シグナルと共にCo-stimulationシグナルによってポジティブにネガティブに制御されている。CD28を介してポジティブなCo-stimulationが行われるのに対して、CTLA-4を介するシグナルは活性化を抑制する。CTLA-4はCD28と同一のリガンド(CD80,CD86)と高親和性で結合し、CD28が常に細胞表面に発現しているのに対して、CTLA-4はT細胞活性化に伴って一過性にのみ発現する。抗CTLA-4抗体を投与することによって腫瘍の生着が抑制されることから、CTLA-4の発現調節と抑制性シグナルの制御は抗腫瘍免疫の活性化に重要である。我々は、CTLA-4の細胞内領域に会合する分子を酵母2ハイブリッド法でクローニングした結果、クラスリン小胞を形成するアダプター複合体AP-2の中μ2とCTLA-4がその細胞内領域のチロシンモチーフを介して会合することを明らかにした。このチロシンモチーフとμ2との結合によってCTLA-4のinternalizationが誘導されることを示した。更に、チロシンモチーフがチロシンリン酸化を受けることによってこの会合が阻害されることが判明した。リン酸化チロシンにはSHP-2がSH2領域によって会合してネガティブシグナルを伝達することが知られているので、CTLA-4はT細胞が活性化され発現しても、リン酸化を受けないうちはAP-2と会合して直ちにinternalizeされ、ほとんど細胞表面発現しないが、リン酸化されるとAP-2とは会合できず細胞表面に留まり、しかもSHP-2との会合を介してネガティブなシグナル伝達を行っていると考えられる。即ち、単一のチロシンのリン酸化によってinternalizationか、シグナル伝達かが制御されていることが明らかになった。
チロシン変異分子をレトロウイルスで導入したT細胞がCTLA-4を高発現することから、この制御機構が正常のT細胞でも行われていることが判明した。また更に、CTLA-4のリン酸化を行うチロシンキナーゼを同定するため、種々のチロシンキナーゼとCTLA-4をco-transfectionする系で解析した結果、srcキナーゼFynとLckがCTLA-4と直接会合してリン酸化することが判明した。
