細胞周期と細胞死のシグナル伝達阻害剤の開発とその分子作用機構

研究概要	ヒストンアセチル化による核内情報伝達、Ras/MAPキナーゼ系、蛋白質核移行系等を介した増殖と細胞死の細胞内情報伝達系を標的とし、これらを正と負に制御する薬剤の開発を行うことを目的とした研究を行い、以下の成果を得た。 (1) 抗腫瘍活性が認められ、その作用機構の不明な化合物の中から、ヒストンアセチル化・脱アセチル化にかかわる阻害剤を探索したところ、がん遺伝子ras等でトランスフォームした細胞の形態を正常化する物質として報告されていた環状デプシペプチドFR901228とJun Dの発現誘導を介してがん細胞の正常化を引き起こしていると考えられていたスルホン鎖アミド系合成化合物Oxamflatinがともに強力なヒストン脱アセチル化酵素阻害剤であることを見出した。特にFR901228はその強力な抗がん活性から、現在米国で臨床試験が進行中であり、ヒストン脱アセチル化酵素の抗がん剤の標的としての重要性が明らかになった。 (2) 酵母を用いたMAPキナーゼホスファターゼの探索系から、活性物質を生産する微生物を見出し、活性物質の単離・精製を行った。現在構造解析中である。 (3) 昨年度、レプトマイシンの標的として同定したヒトCRM1が蛋白質核外移行シグナル(NES)の受容体であることを明らかにした。本年度は、レプトマイシンを用いて様々な制御蛋白質が核外輸送を受ける可能性を検討し、分裂酵母のAP-1様転写囚子PaplのC末端にNESが存在し、CRM1によって核外輸送されることによって機能が阻害されていることを明らかにした。さらに、このNESがある種の酸化ストレスに応答する新規のNESであり、このようなNESを介した酸化ストレス応答が酵母からヒトまで保存されていることを証明した。
