分泌蛋白質特異的Secチャネル複合体の構造・機能

研究概要

大腸菌における分泌蛋白質輸送装置は、SecYとSecEが形成するチャネルに、SecAが挿入・脱離を繰り返して進行する。この時SecGは、その膜内配向性の反転と復帰を繰り返すことによりSecAの挿入・脱離を促進する。SecAやSecGの構造変化の詳細と、因子間の相互作用を明らかにするため、SecG分子内の様々な位置にシスライン残基を一個導入した変異体を多数作製した。これらの変異体SecGはすべて機能を持っており、SecG遺伝子破壊株で発現させると低温での生育が回復した。そこで、変異体を発現した大腸菌から、反転膜小胞を調製し、SH基特異的化学修飾試薬や化学架橋試薬を用いてSecGの膜内での存在様式、他因子との相互作用様式を調べ、以下の成果を得た。
1.SecGの膜内配向性は、これまで提唱してきたモデルが基本的に正しく、N末端、C末端をいずれもペリプラズム側に露出し膜を2回貫通し、膜貫通領域の間の領域は細胞質側に露出している。
2.SecAと相互作用する領域は細胞質側に露出した領域である。
3.チャネル構成蛋白質SecYとは、細胞質側に露出した領域から、2番目の膜貫通領域である。
