神経系におけるグリア細胞の機能の分子遺伝学的解析

研究概要

ショウジョウバエにおけるグリア細胞の分化決定機構を明らかにすることを目的として、グリア細胞特異的転写因子Repo蛋白質の転写制御について解析を行った。ルシフェラーゼアッセイの結果、Repo蛋白質は転写を促進することにより下流標的因子を調節していることが明らかとなった。また、酵母の転写因子であるGal4の認識配列(UAS)をrepo cDNAの上流に有したトランスジェニックフライを作成し、全てのニューロブラストでGal4を発現させる系統と交配することにより repo 遺伝子産物を異所的に発現させた。その結果、異所的なM84(グリア細胞特異的なエンハンサー・トラップ系統)発現細胞が認められた。加えて免疫組織染色の結果、長軸索路の著しい断裂と運動神経の走行異常を認めた。また、repo 遺伝子産物をmidlineの細胞にて異所的に発現させた結果、同様に長軸索路の断裂を観察した。さらに、下流標的因子を単離する目的で、yeastを用いたyeast one-hybrid法によるスクリーニングを行った。3万クローンのスクリーニングの結果、複数の陽性クローンを得た。
