雌雄異株植物を用いた性染色体と生殖器官特異的遺伝子群に関する分子細胞学的解析

研究概要

本研究では、性染色体をもちXY型の性決定を行うナデシコ科の雌雄異株植物メランドリウム(Silene latifolia)を材料に用いて、雄性生殖器官特異的に発現する遺伝子と雄ゲノム特異的なY染色体との関連について解析を行っている。今年度の成果は以下の通り。
1.ディファレンシャルスクリーニングにより単離した4種類の雄性生殖器官特異的(Male Reproductive Organ Specific)遺伝子(MROS1-4)のうち、MROS3のゲノミッククローンをinverse-PCRを用いて単離した。約4kbpと3kbpの2種類のクローンが得られた(MROS3AおよびMROS3Bと命名)。両者のORF部分は93.3%の高い相同性を示すが、上流域・下流域は有意な相同性を示さない。従来得られていたcDNAに対応するMROS3Aの転写領域中の3非翻訳領域には、162bpの非自律性トランスポゾン様の挿入配列が認められた。単離したゲノミッククローンをブローブとするマルチカラーFISHを行った結果、MROS3AおよびMROS3B遺伝子は常染色体の末端部にマップされ、性染色体上には存在していないことが明らかになった。
2.メランドリウム根端分裂組織を材料に用いて染色体標本を作製し、UVレーザービームマイクロダイセクションによりY染色体を直接単離した。回収したY染色体1本(0.5pg)をトボイソメラーゼI処理後DOP-PCRにかけ、鎖長200から300bpのY染色体由来DNA断片を増幅した。増幅されたDNA断片の全体をブローブとしてFISHを行うと、雌のゲノムDNAをブローブの200倍量サブレッサーとして加えてあるにも関わらず、Y染色体特異的なシグナルは検出されず、全ての染色体にシグナルが見られた。このことから、メランドリウムのY染色体はその大部分が他の常染色体やX染色体と共通のDNA配列からなり、特異的な反復配列は蓄積していないと考えられる。また、Y染色体由来DNAのライブラリーからY染色体の末端部分由来である可能性の高いクローンが一つ得られたので、現在解析している。
