カテプシンEの生合成と細胞内輸送および局在化機構の解析

研究概要

本研究では、ラットミクログリア細胞やマウス白血病フレンド細胞、ラット胸腺細胞やカテプシンE遺伝子を導入したCHO細胞といった異なる細胞種を用いてカテプシンEの細胞内輸送と選別機構およびプロセシング機構を分子細胞生物学的に解析した。各細胞に対し[^<35>S]メチオニンを用いたパルスチェイス実験を行い、経時的に細胞と培養上清を回収し、両者に含まれるカテプシンEを特異的なポリクロナール抗体で免疫沈降し、沈降物をSDS-ゲル電気泳動とフルオログラフィーによって解析した。その結果、(1)ミクログリア細胞においては、カテプシンEはプロ型酵素として合成された後、チェイス4時間後には中間体を経て成熟型分子へと変換することがわかった。この間、糖鎖は高マンノース型から複合型へと修飾された。このプロセシングはプロトンポンプの阻害剤であるバフィロマイシンA1によって強く阻害された。免疫電顕による観察で、本酵素の大部分がエンドソーム様の空胞構造に存在していることが確認されたことから、本酵素は最終的にはエンドソーム様の酸性コンパートメントで成熟型酵素への変換が起こっていることがわかった。(2)胸腺細胞では、カテプシンEは複合型糖鎖をもつ中間型までプロセスされるが、成熟型酵素への変換はチェイス72時間後でも観察されなかった。しかし、デキサメサゾンを投与すると、胸腺の萎縮に伴い成熟型カテプシンEが強く観察された。免疫電顕による観察では、正常な胸腺細胞のカテプシンEはゴルジ体および小胞体に認められるが、デキサメサゾン処理によってかなりの部分がエンドソーム様オルガネラに移行することがわかった。(3)フレンド細胞やカテプシンE遺伝子を導入したCHO細胞では、本酵素は高マンノース型糖鎖をもつプロ型分子として大部分が小胞体に存在していた。これらの結果から、カテプシンEは細胞特異的なプロセシングを受けて、それぞれの最終局在部位に輸送されることがわかった。
