新規抗マラリア剤アルテミシニン及びキニ-ネ類縁体の完全合成と作用機構の解明

研究概要	(1)ホモプロトベルベリン誘導体:プロトベルベリンアルカロイドの生合成機構を範として、フェネチルイソキノリン誘導体のN-オキシドから誘導した7員環構造を持つホモプロトベルベリン体は熱帯熱マラリア原虫に対してEC_<50>6.8×10^<-7>Mの活性を示し、毒性比も10倍以上であった。そこで数種の誘導体を新規に合成し、活性を検討している。なお、前記化合物とプロトベルベリン誘導体とを比較することは、活性と置換基およびコンホメーションとの関連を明らかにする上で興味深い。 (2)ビシクロ[6.4.0]ドデカン誘導体:スルホンに対して分子内ミカエル反応に引き続いてのアルキル化、脱離反応をone potで行って得た8員環化合物は熱帯熱マラリア原虫に対してEC_<50>1.9×10^<-8>Mの活性を示した。そこで関連化合物について活性を検討した。 (3)アルテミシニン誘導体:アルテミシニンおよびその類縁体の合成ルートを開発する目的でトリエンの分子内ディールス・アルダー反応を行い、鍵合成中間体となる双環性化合物を収率良く得ることができた。現在、一重項酸素酸化反応等によって、エンドパーオキシド部を構築し、アルテミシニン様の抗マラリア活性が期待される化合物への誘導を検討中である。また、全合成研究と同時に、キク科植物のクソニンジンから、アルテミシニン酸を単離し、現在アルテミシニン誘導体への変換を行っている。上記以外に、分子内二重ミカエル反応、間接電解反応等を利用して、複雑な多環性化合物を数多く合成しており、これらの中から有効な抗マラリア活性を示すリ-ド化合物の発見が期待される。
