胸腺外分化T細胞によるマラリア感染抵抗性の分子機構

研究概要	肝類洞に見い出される胸腺外分化T細胞は、加齢、細胞内寄生細菌感染、担癌状態、及び自己免疫疾患等で著しく活性化してくる。この胸腺外分化T細胞は、自己応答性を持つ禁止クローンを主体としていることが明らかとなっている。すなわち、この胸腺外分化経路は異常自己の監視システムを担っている。種々の病態において、胸腺由来のT細胞では説明できない現象が認められることから、マウスマラリア感染における肝T細胞を主体とする胸腺外分化T細胞の役割を解析した。マウスマラリアP.yoelii17X(致死株)及びP.yoelii17XNL(非致死株)をC57BL/6及びC3H/Heマウスに接種し、肝臓、脾およびその他のリンパ系器官よりリンパ球を分離し、フローサイトメトリーを用いて細胞表面マーカーを解析した。マウスの胸腺外分化T細胞(intTCR細胞)は、CD3(TCRαβ)の蛍光強度が弱く、IL-2Rβ鎖が陽性であることから容易に同定される(CD3^<int>IL-2Rβ^+T細胞)。マウスマラリア感染においては、肝や脾に著しいリンパ球増多が起き、特に肝の腺腺外分化T細胞が増加する。非致死株感染マウスの回復期には、γδT細胞が肝臓で著しく増加する。胸腺外分化T細胞おいては、両株感染とも急性期にはCD4^+細胞が、非致死株の回復期にはCD4^-CD8^-DN細胞が増加している。胸腺外分化T細胞に存在する自己応答性の禁止クローン(Vβ3^+、Vβ11^+細胞)は、非致死株感染の急性期に増加した。感染マウスの肝リンパ球は、感染赤血球に対して細胞障害活性を示した。また、マラリア患者の末梢血リンパ球の解析では、マウスと同様にヒト胸腺外分化T細胞であるCD56^+T、CD57^+T細胞が有意に増加していた。以上の結果は、細胞内寄生であるマラリア原虫感染では胸腺外分化T細胞の活性化が主要な免疫反応であり、しかもマラリア感染自己細胞を異常自己と認識して免疫系が働いていることを示唆している。
