研究概要

本研究では、独自の生体機能探索分子を設計・合成し、膜受容体から核内に至る一連の細胞情報伝達機構の解明に取り組み、いくつかの成果をあげることができた,。鈴木は、脳実質内の新規受容体に強くかつ高選択的に結合する探索分子15R-TICの開発に成功し、脳内に新たなPGI_2受容体サブタイプ(IP_2)が発現していることを明らかにした。さらに、15R-TICのトリチウム標識体を用いたラット脳凍結断片のオートラジオグラフィーにより中枢型IP_2受容体の局所分布の視覚化に成功し、IP_2が情動行動、嗅覚、記憶、学習、痛覚、聴覚を掌る部位に局在することを示した。また、金属触媒反応による高速メチル化反応を開発して15R-TICの^<11>C含有PETトレーサーの合成に成功し、これを用いて生きた赤毛ザルの脳PET画像撮影を行って、IP_2受容体が視床および線条体などの中枢神経系に特異的に分布することを明らかにした。一方、Δ^7-PGA_1メチルエステルおよびその類縁体を活用して、同PGがサイクリンキナーゼ阻害タンパク質p21の特異的誘導とサイクリンEの阻害により細胞周期をG1期で停止させることを明らかにした。柴崎はPKCに強い阻害活性を示すホルボールエステル-ホスファチジルセリンハイブリッド分子に親水性リンカーを結合させた分子を設計し、合成を進めている。また、独自に設計したプローブによるPKCの光アフィニティラベリングを検討し、結合部位を明らかにした。中山はグリベンクラミドの標的タンパク質がスカベンジャー受容体CD36であることを発見し、LDL取り込み抑制作用があることを確認した。また、新規Ca^<2+>拮抗薬の心筋でのCa^<2+>チャネル結合部位を明らかにした。さらに、光アフィニティラベル法での超高感度解析法の開発にも成功した。以上、本年度の研究は概ね計画通りに遂行できた。
