研究概要

本重点領域研究では、飛躍的に進展する細胞生物学、分子生物学及び構造生物学の成果を阻害物質や治療薬の創製などに役立てるため分子生物学者、蛋白構造化学者、有機合成化学者などの研究領域の専門家集団を集めて研究計画が立案されている。4班(第1班:受容体の機能解析と固体レベルでの役割の解明。第2班:細胞内情報伝達の生理機能解析と情報分子の可視化。第3班:プロテインキナーゼを中心とする情報ネットワークの解析。第4班:標的分子の構造情報に基づく分子設計及び有機合成)が置かれ、研究遂行が行われた。本研究の特色は、1から3班の研究課題に関係する探索分子の合成が4班の計画班員によって行われ、作られた機能探索分子は1から3班の研究者にフィードバックされ実験に使用されていることである。本研究を重点的、効率的に発展させ、機動的かつ弾力的な研究班の運営のために置かれた総括班では、各班で得られた成果を合同班会議(平成9年3月1日から3日まで開催)、刊行物(重点ニュース、3回発刊)などを通じて公表し、コンピューターネットワークを導入し情報交換を行った。このような活動は研究成果の社会的還元につながった。更に新規導入した機器による機能探索分子のスクリーニング実験を行った。以上のような活動を通じて、班員の中での有意義な意見の交換と有機的な連絡がとれ、一部の新規合成探索分子は、すでに1から3班の実験に用いられている(抗腫瘍性プロスタグランジンの新規類縁体の創製、特異的探索分子の創製とその活用による脳内プロスタグランジン受容体イメージング、スルホニルウレア剤の結合蛋白質の同定)。以上のように本研究では、生命科学者と化学者の有機的連携がポイントとなっている。来年度も関連諸分野の専門家を結集し、総合的に研究を遂行するために、総括班は異分野の研究者によって構成される各班の有機的、機能的な連絡と運営ならびにそれに付随する機能探索分子の開発実験を行っていく。さらに研究期間中に生ずる新たな発見や進展に迅速に対応するために公募班員の組替えを行う。
