研究概要

蛋白質のリン酸化/脱リン酸化は、多様な生体機能における可逆的な調節を司る重要なメカニズムである。この多彩な役割を演出する受容体/非受容体型、さらにセリン・スレオニン/チロシンキナーゼなど多種類のプロテインキナーゼに関する情報伝達とそのクロストークおよび機能、さらにキナーゼをターゲットとする新規分子プローブの発見を目指した。
受容体型セリン・スレオニンキナーゼであるTGF-βを介した情報伝達に関わる研究としては、TAK1キナーゼの機能探索(松本)とアクチビンの精製および構造生物学的アプローチ(三井)が行われている。さらにTGF-βシグナル伝達を構成する蛋白質として、このシグナル伝達を抑制するSmad6および7が川畑らの研究によって同定されている。非受容体型セリン・スレオニンキナーゼでは、Ca^<2+>/カルモデュリン依存性プロテインキナーゼII(CaMkinase II)αとβアイソフォームがキナーゼカスケードを形成すること、(日高)神経突起伸長を促進することおよびプロテインキナーゼCによってアイソフォーム特異的に調節される可能性が示された(山内)。さらに、small G-proteinであるRhoがミオシン軽鎖をリン酸化したり、ミオシン軽鎖の脱リン酸化を阻害したりして平滑筋の収縮に関与することも見いだされた(黒田)。Rhoによって活性化されるRho kinaseの基質としては、そのほかにERMファミリーとadducinが発見されている(黒田)。またPKCαトランスジェニックマウスが免疫系老化のモデル動物であることも見出され(中島)、さらに骨髄球系細胞増殖・分化の制御におけるCa^<2+>/カルモデュリン(CaM)依存性蛋白Ser/Thrリン酸化反応の役割も明らかにされた(西川・片山)。
受容体型チロシンキナーゼについては、その下流でシグナル伝達を行ういくつかの蛋白質/キナーゼについて研究が進められ、Shcのリン酸化はEGFによるJNKの活性化及び細胞増殖に重要であること(後藤)、PI3キナーゼがRasの活性化を高めること(植木)、さらにMAPキナーゼはPDEキナーゼ活性化に関与しないことが明らかになった(牧野)。非受容体型チロシンキナーゼとしては、SykによるcJun-N-terminal kinase(JNK)の活性化が酸化的ストレスの誘導をおこすことが明らかになった(山村)。また、細胞内チロシンキナーゼBtk・Fyn・LynがCD38を介したB細胞活性化に関与することが明らかにされている。さらにSrcについては塩化第二水銀やNOによる酸化還元反応によって活性化を受け重金属イオンによって起動するアポトーシス誘導性シグナルのカスケードの一端を担うことが判明した(浜口、中島)。野澤はチロシンキナーゼとPLDの間にクロストークが存在することを発見している。
これらの研究は、新規細胞機能探索分子の開発にも大きく貢献している。中西は合成はプロスタグランジンA1(PGA1)アナログの抗腫瘍効果が、p21Cdk阻害蛋白質の発現を誘導によるcyclin A/Cdk2の阻害とcyclin E発現制御によるcyclin E/Cdk2活性の抑制に起因することを明らかにし、より強力な抗腫瘍作用をもつPGA1アナログの開発に取り組んでいる。松本はRafまたはTAK1を発現している酵母を利用して、inhibitorのスクリーニングを行い、radicicolが阻害剤であることを見いだした。遠藤はダントロレンの誘導体によるCa^<2+>放出モードを観察しており、小堤は、アポトーシス誘導型免疫抑制剤ISP-1の作用点が酵母由来のプロテインキナーゼ様蛋白質であることを突き止めている。
