脳グリア・ニューロン受容体シグナリング系応答の解析と機能探索分子の設計

研究概要	脳において各種ストレス(虚血、細菌感染など)に伴い産生されるNOが、各種の病態の発症に関わると示唆されている。脳NOの産生とそのニューロン毒作用を薬理学的に制御することを目的として、脳組織におけるNO産生機構とNOのニューロン死惹起機構を検討した。ラット初代培養グリア細胞にLPSを作用させると誘導型NO合成酵素(iNOS)が誘導され、NOを産生することが分かった。つぎに、二層式ディッシュの上層に初代培養グリア細胞を、下層にNG108-15細胞を培養し、培養液にLPSを加えて作用させたところ、NG108-15細胞においてのみDNA断片化を起こした。sodium nitroprusside(SNP,NO供給薬)もまたNG108-15細胞に対し、DNA断片化を引き起こし、また、そのGAPDH活性を抑制した。NOはグアニル酸シクラーゼを活性化する他に、glyceraldehyde-3-phosphate dehydrogenase(GAPDH)などの蛋白質に対し修飾反応(ADP-リボシル化)を亢進させることが知られている。つぎに、このGAPDH活性低下がニューロン死に関与しているかを選択的GAPDH阻害薬koningicacid(KA)を用いて検討した。KAはGAPDHへの〔^<32>P〕NAD結合、〔^<32>P〕ADP-リボシル化を促進し、また、KAはSNPによるGAPDHへの修飾反応を抑制したことから、KAはNOと同じ作用部位、あるいは極めて近傍に作用することが推定された。さらに、KAをNG108-15細胞に作用させたところ、GAPDH活性を抑制するとともにクロマチンの凝縮とDNA断片化を引き起こした。このことから、GAPDHへの作用(活性低下、あるいは修飾反応)がNOによる細胞死惹起機構の一部に関与していることが示唆された。これらの結果から、1)脳ニューロン死にNOが関与すること、2)脳でのNOの産生抑制とNOのニューロン死を制御する薬物が脳神経変性疾患に有効であること、さらに、3)GAPDHがニューロン死制御薬創製の標的分子として可能性があること、などが示唆された。
