SH基修飾性化学物質を探索子とするストレスシグナル伝達カスケードの解析

研究概要	1重金属イオンやNO遊離物質によるSrcキナーゼの化学修飾部位がSH基であり、少なくとも一部はS-SまたはS-Hg-S結合による分子凝集によって分子の立体構造が変異することでキナーゼ活性が調節されること、この調節は従来から知られるY527の脱リン酸化とは独立の機序でおこることを明らかにした。 2重金属イオンやNO遊離物質による酸化ストレスによって細胞増殖あるいは細胞死が誘導されることを示し、これに至る細胞内シグナル伝達のカスケードに、Srcファミリーのチロシンキナーゼの活性化に続いて、MAPファミリーのセリンスレオニンキナーゼであるERKやJNKおよび転写因子であるc-Junのリン酸化と活性化が含まれることを明らかにした。 3SH基反応性の重金属関連物質をマウス個体に注射し、この個体から取り出したリンパ球について調べた結果、細胞内蛋白質のチロシンリン酸化が増強すること、これに伴って抗DNA抗体などの自己抗体がつくられるようになることを示した。この成績は試験管内で認められた酸化ストレスによる細胞内情報伝達の促進が個体内でもおこること、そのことが個体の病態発生に係わる可能性があることを実験的に示したものである。 4遺伝子修飾動物における導入遺伝子の過剰発現を制御する生体の機構が破綻する機序に酸化ストレスがどのように関与するかを解明するための第1歩として、筆者らが先に樹立し維持しているCキナーゼトランスジェニックマウスの基礎的な性状を調べた。その結果、このマウスでは加齢による免疫系の老化が促進されて現われること、そのプロセス酸化にストレスが係わる可能性があることを明らかにした。
