研究概要

本研究計画は葉と根及びそれらの器官間の物質輸送に携わる道管と師管からなる維管束系のコミュニティーを対象に、同化機能を制御する情報物質の検知、伝達、及びこれらの器官間の輸送プロセスを解析し、以下の成果を得た。
1.情報伝達物質による光合成機能制御の研究
杉山はトウモロコシ葉でサイトカイニンにより誘導されるバクテリア2成分情報伝達系のレスポンスレギュレーターの類緑体遺伝子ZmCip1を同定し、一次構造を決定するとともに、この遺伝子がトウモロコシにおいてサイトカイニンで媒介される無機窒素シグナルの情報伝達に関わることを明らかにした。水野はシロイヌナズナの根で発現する5種のレスポンスレギュレータ(ARR)を同定し、その一次構造を決定するとともに、このうち3種類について大腸菌を用いて精製し、ARRがHis-Aspリン酸転移情報伝達に重要な酵素活性をもつことを明らかにした。佐野は、コムギの栄養及びサイトカイニンに応答し発現誘導されるプロテインキナーゼ類緑体遺伝子wpk4を大腸菌で発現させ、この遺伝子産物が自己リン酸化能をもつことを立証するとともに、このファミリーに属すると考えられる類縁遺伝子をイネからクローニングした。
2.道管・師管を輸送される物質の同定と情報機能ならびに物質輸送制御の研究
林はイネ師管を転流する窒素化合物と移動と代謝回転の速さを明らかにすると共に、多量に転流するチオレドキシンはhはその遺伝子発現及び形質転換体を駆使して伴細胞で合成され師管内に原形質連絡を通して輸送されることを明らかにした。山谷はグルタミンの輸送に関するイネのNADH-GOGATについてゲノム遺伝子構造を明らかにすると共に、そのアンチセンスRNA法によりこの遺伝子産物の発現量が低下した形質転換イネを獲得した。
