研究概要	四肢腱は肢芽間充織に由来し、骨、筋との密接な関連性を持った構築形成過程では、これらの組織との相互作用と、位置特性の指定が必要とされる。腱形成に関わる転写調節因子Six2の発現調節機構について解析を行い、軟骨、表皮からのシグナルが必須であり、Hoxa-13が自脚特異的にこれらの因子に対する感受性を付与していることが明らかとなった。(黒岩) 水晶体分化開始の指標であるδ-クリスタリン遺伝子の活性化には、2つの転写制御因子SOX2とδEF3の共同作用が必要であるが、δEF3はPax6であることが判明した。水晶体分化過程では、Pax6を発現する外胚葉領域の中にSOX2/3発現が誘導され、ついでPax6はSOX2と複合体を形成して水晶体分化に関わる遺伝子群を活性化する事が明らかとなった。(近藤) マボヤotx遺伝子の発現制御領域の解析を行い、前方神経系での発現をドライブする複数の領域を特定した。ホヤemx相同遺伝子の中枢神経系での発現を調べたところ中枢神経系では発現していなかった。ホヤの進化の過程で、中枢神経系での発現を失った可能性に加え、節足動物と脊椎動物の進化の過程で独立に中枢神経系での発現を獲得した可能性を指摘した。(西駕) Hox-c4標的遺伝子mmab1及びそのホモログmmab2KOマウスを作製した。両者ともヘテロ個体は正常だが、mmab1ホモマウスは眼、生殖器、胸骨の形成異常を示した。眼、生殖器はmmab2と発現が重複しない領域であり、重複する領域では異常は観察されなかった。胸骨はHox-c4KOマウスでも異常が観察され、胸骨形成過程でHox-c4によるmmab1の発現制御が示唆される。(高橋) ショウジョウバエのstill life遺伝子のマウスホモログ,STEFはRacに特異的なグアニンヌクレオチド交換因子として機能している。ドミナントネガティブ型遺伝子を用いた実験からSTEF分子がRacの活性化を介してアクチン細胞骨格系の構造変化を引き起こし、神経細胞の移動、突起伸展、シナプス形成に関与していることが示唆された。(鍋島) ショウジョウバエの発育異常を示す突然変異の解析から、個体のサイズを決定する原因遺伝子が進化的に保存され、多機能未知のタンパク質をコードすることを見い出した。強制発現系での表現型に対する遺伝的相互作用の解析から、そのE2Fの上流での機能と、細胞の発育に関わるインスリン受容体シグナル伝達系への関与が示唆された。(西田)
