枯草菌蛋白質分泌系で作用するSRP様粒子のシャペロンとしての機能解析

研究概要	シグナル認識粒子(SRP)はイヌやヒトのすい臓細胞を中心に研究され、真核生物細胞の蛋白質分泌過程において中心的な役割を荷っている。ヒトSRPは1分子のRNA(SRP 7S PNA,300塩基よりなる)と6種類の蛋白質から構成される。また大腸菌における蛋白質分泌は主にSec蛋白質群によると考えられている。しかし大腸菌にもSRP 7S RNAの相同RNAとして4.5S RNA(114塩基からなる)が、またSRP54蛋白質の相同因子としてFfh蛋白質が見いだされている。しかしその機能はまだ充分には明かになっていない。一方枯草菌は大腸菌と異なり、培地中に大量の蛋白質を分泌生産し、胞子を形成するグラム陽性細菌である。私共は枯草菌にはSRP系とSec分泌系の両方が存在し、これらが共同作業することによって非常に効率の良い蛋白質分泌系を形成している可能性を示し、そのカスケードを研究してきた。枯草菌SRP様粒子はsmall cyto plasmic RNA(271塩基からなる)とFfh蛋白質以外に分子量的10kDaのHBsu蛋白質(大腸菌DNA結蛋白質HU相同物質)がscRNAのドメインに結合していることを明らかにした。枯草菌SRP様粒子はRNA成分の大きさからも推定されるようにヒトやイヌのSRPと類似点が多い。枯草菌に存在するSRP様粒子が翻訳修了後の分泌蛋白質前駆体と複合体を形成するためのシャペロンとして非常に重要な働きをしていることを明らかにした。一方ほ乳類SRPおよび大腸菌Ffhは翻訳途中の前駆体と結合すると考えられている。また枯草菌には大腸菌に存在するSecBは検出されず、一般のシャペロン蛋白質DnaKが蛋白質の分泌系には作用しないことを明かにしている。また枯草菌染色体DNA全塩基配列を決定し、Nature誌に発表しているので新しい遺伝子の解析は容易である。
