研究概要

(A)αBクリスタリンを恒常的に高いレベルで発現しているヒトグリオーマ細胞株U373MG細胞を用いてαBクリスタリンのリン酸化について解析し、以下の結果を得た。1)αBクリスタリンもhsp27と同様に、種々の化学ストレス(亜砒酸ナトリウム、ホルボールエステル、過酸化水素、ソルビトールやNaCl添加による高張ストレス、アニソマイシン、オカダ酸)や熱ストレスによって最高三カ所のセリン残基がリン酸化を受ける。2)その位置はSer19,Ser45およびSer59である。3)これらのリン酸化は環状AMP依存性プロテインキナーゼに依らず、p38とp44/42MAPキナーゼカスケードに関与する酵素によって触媒される。4)αBクリスタリンのストレスによるリン酸化は、U373MG細胞のみならず、C6ラットグリオーマ、3Y-1ラット線維芽細胞でも観察される。5)ストレスとは別に、細胞分裂期にSer45とSer19のリン酸化が特異的、普遍的(調べたすべての培養細胞株およびマウス胎児の組織切片)に高進していることを、それぞれのリン酸化を特異的に検出する抗体を用いて明らかにした。
(B)血管が環状ヌクレオチド依存性に弛緩する際に、約20kDaの二つの蛋白質がリン酸化されることが分かっていたが、これらの蛋白質が、我々の発見したp20のリン酸化されたものであることを明らかにした。
