研究概要	アデノウイルスE1A蛋白質及びレチノイン酸(RA)による胚性腫瘍細胞F9の分化誘導機構を解析する目的で、発癌遺伝子c-junのトランス活性化機構に焦点をあて研究をすすめている。c-junの強制発現はF9細胞を間葉細胞に分化誘導させる事により、c-junのトランス活性化に必要なエレメントDRE(Differentiation Response Element)をc-junプロモーターの-190〜-46に見いだし、これに結合する核内因子DRF(Differentiation Response factor)-1,-2を同定した。E1A及びRAによる分化に伴いDRF-2は消失し、DRF-1は移動度が少し遅いDRF-1'となる。E1Aのドメイン消失変異株、ゲルシフト、プロモーター活性の解析によりDRFは分化に伴いリン酸化される事を見いだした(EMBO.J.14.3496-3509,1995)。本年度は、このDRF複合体にアデノウイルスE1A結合蛋白質でコアクチベータ-であるp300及びATF-2転写因子が含まれる事を初めて明らかにした。相互の免疫沈降反応、Tag付発現ベクターによる相互の免疫反応、in vitroにおけるGST-pull downアッセイや、バキュロウイルス蛋白質を使用した解析によりp300のBr-C/H2ドメイン(963-1571a.a)とATF-2の(112-350a.a)のドメインが直接会合する事、及びATF-2の121のSerineのPKCαによるリン酸化がATF-2/p300によるトランス活性化には必要である事を証明した(Genes & Dev.12,233-245,1998)。今後はp300,ATF-2以外の第三の分子の同定が急務である。その他、p300とCBPの分化誘導、G1アレスト、アポトーシスの機能分担をリボザイムやアンチセンス法を用いて解析を進めている。
