研究概要

平成12年度の主要な研究成果を以下に整理して述べる。
1.翻訳調節におけるRNA情報機能の研究:(1)遺伝暗号の中で最後まで解読のメカニズムが明かにされなかった終止コドンが、解離因子のペプチド・アンチドンにより解読されることが明らかにされた。ペブチド・アンチドンの発見は歴史的な業績であり、さらに解離因子のリボソームへの結合様式がラヂカルフットプリント法により詳細に解析され、tRNA擬態性がトポロジーの上からも証明された。(2)高度好熱菌由来のリボソーム再生因子RRFの2.6A結晶構造が明らかにされ、tRNA構造との擬態性を明らかにした。(3)最近発見されたアンチザイムのファミリーメンバー(AZ2,AZ3)に関するフレームシフト特性の検討により、シフト部位上流および下流の信号配列双方の重要性を明らかにした。(4)HCVの3'非翻訳領域中に存在するポリU領域が、RNAの安定性とは無関係にHCV IRES活性を抑制することを明らかにした。
2.RNAプロセシング機構の研究:(1)大腸菌LacImRNAがSsrA RNAによるトランストランスレーションの特異的なnatural targetであること、LacIのtaggingとproteolysisがlacオペロンの速やかな誘導に寄与していることを見いだした。(2)神経特異的RNA結合蛋白質であるHu蛋白質群が神経分化誘導能をもつことを発見し、その機能ドメインを明らかにした。
3.RNA情報構造機能の研究:ショウジョウバエの性決定に係わるSxl蛋白質とその認識配列RNAとの複合体の結晶構造が解かれた。二つのRNA結合ドメインが認識配列を特異的に認識するその仕組は、これまで知られていないRNA/蛋白質の分子認識のメカニズムを明かにしたもので重要な成果である。また、HuCのRNA結合ドメインの立体構造とRNA認識機構を明らかにした。
