生物分子モーターの動的構造解析

研究概要	本年度の「生物分子モーターの動的構造」班の成果は次のようなものである。1)ミオシンモータードメインの構造変化を1分子レベルで追跡するためのsingle-pair FRET(fluorescence resonance energy transfer)法の開発。1分子のミオシンモータードメインの2箇所を特異的な蛍光色素で標識し、標識分子ひとつから出る蛍光スペクトルを分光し、記録するための全反射照明顕微鏡を製作した。これを用いて、1分子のモータードメインのレバーアームスイングを可視化することを試みた。スイング前の状態とスイング後の状態に固定された変異ミオシンモータードメインをもちいてsp FRETを観察したところ、明らかにスペクトルに変化がおこり、スイングがおこっていることが確認できた。今後は、ATP加水分解の時間経過とレバーアームスイングの時間経過との相関をとる実験をおこなう予定である。2)「新たな生物分子モーターの探索」班でクローニングした粘菌細胞の新規ミオシン、MyoMとMyoKはProcessiveモーターである可能性が強い。これを確かめる実験のため、これら新規ミオシンの発現系構築をおこなった。3)ダイニンの構造機能相関をあきらかにするため、組換えダイニンを発現させる系を構築した。このため細胞性粘菌からダイニン重鎖モータードメイン部分をクローニングし、粘菌発現ベクターに組み込み、粘菌細胞で発現させた。粘菌細胞内ではSDS電気泳動で十分確認できる量の、組換えダイニンモータードメインが発現していた。この組換えダイニンモーターの部分精製をおこなった。今後は、この大量発現系をもちいて、結晶化にもちいることができる量のダイニンモータードメインを精製する予定である。
