神経細胞死とアポリポ蛋白受容体について

研究概要	アポリポ蛋白E(ε4)がアルツハイマー病発症の危険因子とされている。しかし、その遺伝子産物であるApoE4が、アルツハイマー病の病態機序にどのように係わっているかは不明である。そこで、ApoEのイソ蛋白の違いが神経細胞死に関連する可能性を想定し、実験を行った。 ApoEは細胞膜に存在するApoE受容体を介して取り込まれることが知られている。そこで神経細胞に発現しているApoE受容体(LDL受容体、VLDL受容体、apoE2受容体等)の受容体を強制発現するCHO細胞を作製し、この培養細胞系を用いてApoEイソ蛋白の細胞毒性をMIT法あるいはWST法により検討した。3種のヒトApoE受容体cDNAを発現するネオマイシン耐性のCHO細胞を得た(CHO/LDLR、CHO/VLDLR、CHO/apoER2)。これらの細胞の培養系にリコンビナントApoE2,3,4(150mM)を添加し、継時的に抗ApoE抗体による蛍光抗体法で細胞内のApoE動態を検索した。いずれの細胞においてもApoE3,4では添加後30分頃より細胞質内に特異蛍光を認め、その強度は3時間後まで増強していった。それ以降、特異蛍光は細胞の辺縁に局在するようになり、やがて消退していった。ApoE3,4を取り込んだ細胞は核が肥大し、かつプラスチック皿より剥離する傾向を示した。ApoE2を添加した場合は、どの細胞も微弱な蛍光しか発さず、また形態変化も認めなかった。ApoEを添加後3時間の時点でMIT法により細胞毒性を調べると、どの細胞においてもApoE4>3>2の順に細胞毒性効果を認めた。次にH_2O_2の毒性とApoEの効果との相関を調べるために、これらの細胞でまずH_2O_2の細胞毒性を調べた。0mM〜0.5mMまでの濃度では、用量依存的な細胞毒性が認められた。そこで、0.25mM濃度のH_2O_2の細胞毒性状態において、ApoEイソ蛋白の効果を調べたところ、ApoE4>3>2の順にH_2O_2毒性を助長する傾向が認められた。ApoE2については、その取り込み効率が低いので評価はできないが、ApoE3とApoE4を比較した場合、ApoE4の方が細胞内からの細胞毒性効果が強いことが明らかとなった。
