CAGリピート伸長による、神経細胞死の組織特異性の解析

研究概要	(1)DRPLA遺伝子の発現調節を解析するために、まず5'上流域の塩基配列を決定したところ、すでに報告されているHD遺伝子の上流域と極めて類似していた。そこでDRPLAおよびHDについて、相同領域のプロモーター活性を測定して比較し、転写調節因子や薬剤に対する効果に違いを見出した。 (2)アポトーシス過程におけるDRPLA蛋白質の限定分解 CAGリピート伸長病における神経細胞死とアポトーシスの関係が示唆されている。アポトーシス過程でDRPLA蛋白質は限定分解を受けることを見出し、caspase-3の関与を推定し、切断部位を同定した。さらにDRPLA蛋白質の細胞内局在化と、アポトーシス切断後の細胞内局在化を解析した。 (3)DRPLA遺伝子の機能 DRPLAと相同性の高いアミノ酸配列を有する、EST-M78755で代表される遺伝子の単離を進め、また、DRPLA蛋白質と会合できる蛋白質の分離を進めている。
