研究概要	本研究は平滑筋の分化と形質変換の分子機構解明を研究課題としている。動脈硬化では平滑筋細胞は平滑筋特異的遺伝子発現を消失し、脱分化平滑筋が増殖するが、その機序の解明は動脈硬化の病態理解と対策の構築に必須である。そこで(1)平滑筋特異的転写調節機構と、平滑筋が合成型へ形質変換する際の遺伝子転写機構を明らかにする、(2)血管平滑筋の形質変換を制御する増殖ならびに増殖抑制因子の細胞内情報伝達系を明かにする、(3)高効率の遺伝子導入法を用いて、動脈硬化の成因解明と平滑筋遺伝子の血管系における機能を解明し、遺伝子治療の可能性を明らかにすることを目的とした。まず永井らは平滑筋の形質変換にZn finger型転写因子BTEB2とホメオ蛋白Hexが関与することが明らかとなった。BTEB2は胎児型平滑筋ミオシンSMembの-100bpのGCboxに結合し、SMembだけでなく、組織因子、iNOS、PAI-1、VEGF等の血管病変で発現する遺伝子を活性化した。BTEB2は増殖因子の遺伝子を活性化する転写因子Egr-1や脱分化平滑筋で活性化される組織因子遺伝子を活性化する。BTEB2遺伝子自体は転写因子Egr-1によって制御されることを示した。平滑筋特異的遺伝子発現に関しては平滑筋αアクチン遺伝子の第一イントロンに存在するCArG boxが重要であった。鍋島らは老化抑制遺伝子klothoは内皮細胞におけるNO産生を高め、平滑筋の形質変換を防止すると考えられた。森下、金田らはHVJリポゾーム法による血管壁への遺伝子導入の効率を高めることに成功した。
