血管平滑筋の細胞周期調節機構とシグナル伝達系との相互作用に関する検討

研究概要

血管平滑筋に存在するGATA結合転写因子のisoformを検討するとともに、その発現調節機構および細胞増殖に対する作用を調べた。1)degenerate primersを用いて増幅したZF domainの塩基配列を調べた所、全てのクローンはGATA-6をコードしていた。Library screeningにより単離されたヒトGATA-6は449個のアミノ酸から構成され、推定分子量45.3KDであった。2)静止期のヒトおよびラット培養血管平滑筋細胞を血清で刺激すると、刺激後4hrをピークにGATA-6 mRNAのレベルは著明に減少した。3)GATA-6は蛋白レベルで血管平滑筋に発現されていた。4)deletion mutantを用いた解析からZF domainおよびそのC末端側の塩基性アミノ酸に富むbasic domainがDNAに結合するために必要な領域であった。5)ラット培養血管平滑筋細胞にtransfectされたGATA-6 wild type(GATA-6wt)はZF domainを欠失mutant(GATA-6DZF)に比べ、BrdUの取り込みを60%抑制した。またp53-/-マウス線維芽細胞においてもGATA-6wtはBrdUの取り込みを95%抑制した。p53-/-マウス線維芽細胞ではGATA-6wtを取り込んだ細胞にp21の強い発現を認め、またp21のpromoterはGATA-6wtにより活性化された。さらにp21-/-マウス線維芽細胞においては、GATA-6wtのBrdU取り込み抑制作用は弱く37%の抑制であった。結論:血管平滑筋に発現されるGATA結合転写因子の主要なisoformはGATA-6であり、血管平滑筋細胞において過剰に発現されたGATA-6は細胞周期のS期進入を抑制し、この作用の少なくとも一部はp53を介さないp21の発現増加による事が示唆された。
