動脈硬化発生・進展における血管内皮細胞機能に関する分子病理学的研究

研究概要

本年度の主な研究業績を以下に示す。
1.動脈硬化発症における内皮細胞の接着因子発現機構:apo(E)欠損マウスにおける動脈硬化早期病変は、shear stressの弱い部に好発しVCAM-1とICAM-1の内皮細胞発現と相関することがin situ免疫電顕法で確認された。特にVCAM-1発現は血中のコレステロール値と関連があることより、動脈硬化発生には内皮細胞のVCAM-1が重要である可能性が示唆された。
2.PTCA後再狭窄の発生および予防のための遺伝子制御:1)HVJ-リポゾーム法でwild-type p53遺伝子を血管平滑筋細胞に導入すると、in situにて細胞周期をG1期に誘導して増殖を抑制し、さらにin vivoではバルーン障害後の内膜肥厚を著明に抑制した。2)CMV-IE遺伝子を家兎総頚動脈に導入すると内膜の細胞線維性肥厚を惹起することから、動脈硬化発生にCMV感染が関与する可能性が示唆された。
3.動脈壁リモデリングにおける内皮細胞機能:1)硬化内膜内の新生血管形成に平滑筋細胞、マクロファージが産出するVEGFが関与していること、2)一方、新生血管の内皮細胞でのTie2発現が減少していることから、硬化内膜内の新生血管形成にはアンジオポイエチン-Tie2連関の抑制が関与している可能性が有ること、3)このVEGF発現亢進には、その遺伝子のプロモーター領域でのAP-1やSp-1結合が低酸素とともに関与していることがこれらdecoy遺伝子を用いた解析より明らかとなった。
