中性化合物の高感度分離同定用ミセル動電クロマトグラフィー・質量分析法の構築

研究概要

本年度も主として3つの研究を平行して進めた。一つはMEKC-MSを行うための準備として,MEKCによる中性試料のオンライン濃縮法についてさらに検討した。昨年度中にスタッキング法により疎水性の強い中性化合物であれば,100倍程度の濃縮が可能となっていた。濃縮機構を検討中に全く新しい原理に基づいた濃縮法であるスウィーピング法を開発できた。疎水性の高い中性化合物では5000倍までのオンライン濃縮が可能となった。ミセルと反対電荷を持つ試料については数十万倍の濃縮も可能であることも明らかになりつつある。これら濃縮法は,化合物選択性があるので,環境試料,生体試料などの特定成分の濃縮に有効になると期待できる。もう一つは本研究で新しく購入した質量分析計であるFinnigan LCQとキャピラリー電気泳動(CE)装置HP 3DCEを用い,質量分析法(CE)-MSシステムの利用して,MEKC-MSの高感度化の検討である。実験を主に担当していた外国人特別研究員が去り,博士課程学生に交代したので,装置の取り扱いに習熟するのに時間がかかり,まだ大きくは進展していない。MS装置にも,CE-MSには不都合な点が明らかになり改良を行った。この研究は次年度に大きく進展できる予定である。上記試料濃縮法と組み合わせて,高感度MEKC-MSシステムを完成する予定である。もう一つは研究協力者の所有する装置を用いたMEKC-MSの研究である。部分注入法,逆移動ミセルを用いる方法,界面活性剤の除去を行わずそのままMEKCを行う等種々の方法を試みたところ,どの方法を用いてもMEKC-MSは可能であった。分離との兼ね合いでどの方法が都合がよいかを決めればよいことが明らかになった。しかし,界面活性剤を除去しない場合,検出感度は他の方法より劣った。
