粘膜免疫における上皮細胞とγδ/αβT細胞間インターネットの解明

研究概要

粘膜免疫機構の基礎的解明、特に病原微生物やアレルゲンの侵入にさらされている粘膜上皮細胞、粘膜系γδ/αβT細胞、B細胞の三者が構成する粘膜細胞間インター・イントラネットについて明らかにすることを目的としている。本年度は、粘膜免疫誘導機構の中枢的役割を果たしている腸管関連リンパ組織(GALT)の代表格であるパイエル板における粘膜細胞間イントラネットを検討した。パイエル板の組織構築に関しては、IL-7RとLTβRを介したシグナルが必須である事が解っている。そこで妊娠マウスをLTβR-Ig複合タンパクで処理するとパイエル板欠損マウスが生まれてくる。この手法によりパイエル板が欠損している事は、解剖学的、組織学的に確認した。次にパイエル板欠損マウスを蛋白抗原(OVA)と粘膜アジュバントとしてコレラ毒素(CT)を混合して経口免疫した。対照群としては、LFA-3-IgとPBSで処理したパイエル板存在マウス群を経口免疫した。経口免疫後にOVA特異的IgA抗体産生を検討すると、各種分泌液中の抗原特異的IgAのレベルは対照群に比較して低下していた。しかしながら、両者群には統計学的にはまったく差がなかった。同様な結果が粘膜関連組織、例えば腸管粘膜固有層から分離した単核球で比較・検討すると同様に抗原特異的IgA抗体産生細胞の頻度に関して実験(パイエル板欠損マウス)群と対照群では顕著な差がなかった。この結果はIgA産生過程においてGALTの代表格であるパイエル板に依存しないIgA誘導システムが存在していることを示唆している。そこで、粘膜免疫の誘導組織(パイエル板)と実効組織(腸管粘膜固有層)におけるB-1細胞とB-2細胞の存在頻度を比較するとパイエル板のB細胞はB-2細胞で構成されていた。一方、後者ではB-1細胞とB-2細胞が両方、存在していた。この結果は、B-2細胞はいわゆる誘導・実効組織からなるCMISに依存し、またB-1細胞はそれとは独立したシステムによって誘導制御されている可能性を示唆している。さらに、B-1細胞とB-2細胞におけるサイトカイン制御機構を検討すると、B-1細胞はIL-5RとIL-15Rを発現し、両レセプターに特異的サイトカインにより細胞の分化増殖が制御されていた。一方、B-2細胞はIL-5RとIL-6Rを発現し、いわゆるTh2型サイトカインにより誘導制御されていた。この様な事実を統合するとIgA誘導系はCMIS依存型(B-2細胞)とCMIS独立型(B-1細胞)が存在し、IL-5/IL-5Rを共通として各々独立したサイトカインの制御(IL-6/IL-6RとIL-15/IL-15R)を受けている事が強く示唆された。この知見は、粘膜細胞間イントラネットを使った粘膜ワクチン開発に向けて新情報を提供した。
