研究概要

1.水チャネルのクローニング
水チャネルAQP9のホモログである水チャネルAQP9Lをクローニングした。mRNAの発現は肝臓で最も多く、精巣にも認められた。機能的には、AQP9Lは水のみならず、グリセロール、尿素にも透過性があることが判明した。
2.水チャネルの構造の解明
水チャネルの中でもAQP2とMIPはアミノ酸相同性がきわめて高い(58%)にもかかわらず、水透過性AQP2で高くMIPで低い。そこでAQP2とMIPのキメラを作製して検討し、細胞膜貫通部のヘリックス5にあるアミノ酸が、高い水透過性に関与していることを見い出した。
3.尿濃縮機構に関わる細胞内メカニズム、細胞内情報伝達系の解明
ラット髄質集合尿細管を高浸透圧に暴露するとNa-K-ATPase活性が低下したが、短時間暴露と長時間暴露では活性低下の機序が異なっていた。その他、LLC-PK1細胞やメサンジウム細胞などを用いた検討で細胞内情報伝達系に関する知見を得た。
4.腎性尿崩症の遺伝子異常の解明
常染色体優性遺伝腎性尿崩症の患者で見い出されたAQP2の変異である809del7が、どのような機序で尿崩症が発症するのかを検討した。ツメガエル卵母細胞を用いた発現実験から、809del7は転写の抑制あるいは蛋白のトラフィッキンング異常が発症原因であることが示唆された。これらの結果は昨年の第32回アメリカ腎臓学会で発表し、現在論文を準備中である。
