HNK-1糖鎖抗原に関する分子生物学的研究

研究概要

代表者らはすでにHNK-1糖鎖抗原の生合成に関わるグルクロン酸転移酵素のcDNAクローニングに成功しこれをGlcAT-Pと名付けている。本年度はGlcAT-P以外のHNK-1糖鎖抗原の生合成に関わる第二のグルクロン酸転移酵素の存在を予測し、ラット脳cDNAライブラリーをGlcAT-PcDNAをプローブとしてlow stringency条件でスクリーニングした結果、第二のグルクロン酸転移酵素のクローニングに成功し、これをGlcAT-Sと名付けた。 GlcAT-Sは324アミノ酸残基よりなるII型の膜貫通タンパク質であり、GlcAT-Pとはアミノ酸レベルで49%のホモロジーを示した。
Northcm blot解析によりGlcAT-S mRNAの分布を調べたところ、大脳・小脳などの神経系にのみ、2.3kbおよび1.1kbのmRNAが発現しており、HNK-1糖鎖抗原の細胞分布とよく一致していた。さらに、GlcAT-S cDNAをCOS-1細胞に強制発現させると、細胞表面にHNK-1糖鎖抗原が発現するととも細胞の形態が変化し、神経細胞様の突起が観察された。これらの結果は、GlcAT-Pについてすでに報告した結果とよく一致しており、GlcAT-SがHNK-1糖鎖抗原の生合成に関わる第二のグルクロン酸転移酵素であることを示している。さらに、使用される糖転移酵素の種類に関わらず、HNK-1糖鎖抗原の発現がシグナルとなり細胞の形態変化を引き起こすという興味ある事実を証明したものである。なお、GlcAT-S cDNAをCOS-1細胞に強制発現させた細胞全体のWestemblotにより、80-208kDaの内在性タンパク質にHNK-1糖鎖抗原の発現が示されており、本酵素はタンパク質性の受容体にグルクロン酸を転移することが確認された。
