マラリアの生殖母体をめぐる伝播阻止の研究

研究概要	マラリア伝播に関わる諸問題をそれぞれの分担研究者の研究対象、方法論に従って取り上げた。マラリア伝播者ハマダラカの媒介の根源である唾液腺に注目しその構成成分を分析した。吸血行動により成分が変動する過程を追跡した。。ハマダラカ唾液の物質の一つのクローニングに成功しネッタイシマカのアレルゲンと30%のホモロジーがあると確かめられた。胞子体(sporozoite)がマラリア伝播者であるハマダラカの中で出入りし排出液(prediuresis)に検出された。ハマダラカの中のネズミマラリアのオーキネートの発育を阻止するマラリア表面分子(Pbs21)に対する抗体を用いて蚊の中で伝播を阻止する試みを行い、単クローン抗体の変異部位をコードする遺伝子をクローニングし配列を決めた。この分子をバキュロウィルスを用いて産生しマウスに投与してハマダラカに吸血させたところオオシストの形成を阻止した。マラリア伝搬の阻止につながる。(松岡、石井) マラリア伝搬のステージである生殖母体を殺減する薬物プリマキンは投与する際、副作用が問題になるG6PD酸素欠損者を検出する新しい簡便法が開発され流行現場で試行した。結果は良好かつ有望でマラリア流行地住民の評価も高かった。(石井、川端)伝播阻止抗原と共にワクチン候補分子の検討、それに伴う免疫学的反応の分析も行い多様性を認めた。IgG subclassの働きに違いが認められた。(太田) 生殖母体形成誘導を試みヘマトクリット値を高めると転換率が上昇することを確認した。株差があり長期間で形成能を失うものもあり何らかの遺伝子の脱落が示唆された。(川本)オオシスト形成に活性化酸素が関与していることを産生能の低いp47 knockoutマウスを用いて確認した。オオシスト形成に働くキチナーゼの検討を行った。(大橋、村主)ネズミマラリアでTRAP(thrombospondin related adhesive protein)に属するCTRPがookinate特異的に発現し中腸に侵入する際の重要な接着タンパクである事を確認した。(鎮西)マラリア伝播が自然界で起きている現象の理論疫学的解析では現地の疫学データが取れたので入力した上、地理情報システムの活用による分析の試みも現地での情報を採集して行った。(川端、石井)
