研究概要	本研究はツツジ植物などの二酸化窒素同化能力の高い高能力株と低能力株を実験材料として、二酸化窒素ガスとの接触で発現する植物遺伝子の単離と形質転換の研究を行うことを目的とする。本年度は以下の成果が得られた。 1)4ppm NO2に8時間暴露したヒラドツツジの葉から抽出したタンパク質をPI4.2-7.0、MW10-150kDの範囲で二次元電気泳動で展開し、銀染色でスポットを検出した。約1200のスポットの強度を比較した結果、PI5.6、MW25-26kD付近に泳動される2つのポリペプチドスポット強度がNO2暴露処理を行った葉で無処理のものと比較して5倍から7倍増加した。 2)これらスポットのアミノ酸配列を気相法により決定した。ホモロジー検索を行ったところ、これらスポットのポリペプチドは、Peachの2種のauxin-binding proteinやArabidopsisなどの4種のgermin-like proteinのN末端領域と57-68%の相同性があった。 3)ヒラドツツジを0.1ppmのNO2で暴露し、0、1、2、4、8時間後の葉から全RNAを抽出し、ディファレンシャルディスプレイ法によりNO2暴露に応答して経時的に増加または減少するmRNAバンドを調査した。200から900bp間に45〜81本のバンドが検出された。 4)このうち約340bpの位置に認められた発現量が経時的に増加するバンドと約280bpと230bpの減少するバンドからcDNAを回収、クローニングし、それらをプローブとしてノザンブロット解析した。その結果、340bpのバンドクローンをプローブとしたとき約1.3kbに明瞭なバンドが認められ、NO2暴露8時間でバンド強度は約5倍に増加した。このクローンの337bpの塩基配列を決定した。ホモロジー検索の結果、このクローンとホモロジーを持つ既知のタンパク質やcDNAはなかった。
